: Mice were housed and handled according to local, state and federal regulations. All animal studies were carried out according to the protocols approved by the Institutional Animal Care and Use Committee at University of Cincinnati (Cincinnati, OH, USA). <i>Tsc1</i><sup>iΔEC</sup> tumour cell line was established by us as described in the Methods section. LN229 and LEF2 tumour cell lines were gifts from other labs as described in the Methods section. LN229 cells (ATCC® CRL-2611<sup>TM</sup>) were obtained from Dr. David R. Plas (University of Cincinnati College of Medicine) as a gift. LEF2 cells were initially obtained as a gift from Dr. Kun-Liang Guan (University of California at San Diego) and as described previously.<sup>29</sup>
