: All experimental protocols were approved the Ethics Committee of the Suzhou Ninth Hospital Affiliated to Soochow University (Suzhou, China; ethics approval code: NL-2023-1). All methods were performed in accordance with the relevant guidelines and regulations. Informed consent was obtained from all subjects and/or their legal guardians.Several limitations of this study should be acknowledged. First, functional experiments were performed in murine ATDC5 cells rather than primary human nucleus pulposus cells. Primary human NPCs are scarce, highly variable between donors, and difficult to culture and transfect stably; ATDC5 cells were used to ensure robust and reproducible functional results. Future studies will validate the regulatory functions of miR-3620-5p/CPNE6 and miR-6511b-3p/B3GAT1 axes in primary human NPCs. Second, the clinical sample size was relatively modest, which may limit statistical power and generalizability. Larger multi-center cohorts are needed to further confirm the diagnostic and prognostic value of these miRNA-mRNA biomarkers.
