Ethics approval and consent to participate: All experiments involving human pluripotent stem cells were approved by the Institutional Review Board of the National Center for Child Health and Development. The human induced pluripotent stem cell (iPSC) lines used in this study were previously established from healthy donor cells under the approved project entitled “Establishment and Characterization of Induced Pluripotent Stem Cells Derived from Healthy Donor Cells” (approval number NCCHD-1126; approved on December 24, 2020). The use of human iPSC- and ES cell–based experimental systems for studies of hematopoietic differentiation was additionally approved under the project entitled “Studies on Bone Marrow and Blood Cell Differentiation from Human ES Cells” (approval number NCCHD-ES#61; approved on June 19, 2024). In compliance with the Declaration of Helsinki, informed consent was obtained from all cell donors. Human cells in this study were utilized in full compliance with the Ethical Guidelines for Medical and Health Research Involving Human Subjects (Ministry of Health, Labor, and Welfare (MHLW), Japan; Ministry of Education, Culture, Sports, Science and Technology (MEXT, Japan) and performed in full compliance with the Ethical Guidelines for Clinical Studies (Ministry of Health, Labor, and Welfare, Japan). All animal experiments were approved by the Institutional Animal Care and Use Committee of the National Center for Child Health and Development under the approved project entitled “Research on Regenerative Medicine and Cell Therapy Using Mesenchymal Stem Cells, Induced Pluripotent Stem Cells, and Embryonic Stem Cells” (approval number: A2003-002; initially approved on February 28, 2011), and were conducted in accordance with the basic guidelines for the conduct of animal experiments under the jurisdiction of the Ministry of Health, Labour and Welfare (Notification No. 0220–1 of February 20, 2015).
